Studies were conducted to evaluate an efficient sampling method for <i>Varroa destructor</i> density in 954 honey bee colonies in 31 apiaries in Minnesota, North Dakota, California and Texas, USA, from 2005 to 2007. Approximately 35 adult honey bees were collected from each frame in each colony and the date, beekeeper, yard, pallet, colony, frame location and comb contents were recorded. The mite density in each adult bee sample was also determined. Using data from the 954 colonies, the mite infestation level, number of adult bees in a colony, colonies in an apiary and mite spatial distribution were also recorded. Results showed that the treatment thresholds for colonies in a stationary apiary was 10-12% colony mite infestation in autumn, while the treatment thresholds for migratory beekeepers varied depending on the region.
