AIM:To identify genes differentially expressed in mouse hepatocarcinoma ascites cell line with low potential of lymphogenous metastasis.METHODS:A subtracted cDNA library of mouse hepatocarcinoma cell line with low potential of lymphogenous metastasis Hca-P and its synogenetic cell line Hca-F with high metastatic potential was constructed by suppression subtracted hybridization (SSH) method. The screened clones of the subtracted library were sequenced and GenBank homology search was performed.RESULTS:Fifteen differentially expressed cDNA fragments of Hca-P were obtained which revealed 8 known genes, 4 expressed sequence tags (ESTs) and 3 cDNAs showed no homology.CONCLUSION:Tumor metastasis is an incident involving multiple genes. SSH is a useful technique to detect differentially expressed genes and an effective method to clone novel genes.

Abscission in plants is tightly controlled by multiple phytohormones and the expression of various genes.However,whether the plant hormone brassinosteroids(BRs)are involved in this process is largely unknown.Here,we found that exogenous application of BRs reduced the ethylene-induced fruitlet abscission of litchi due to lower ethylene(ET)production and suppressed the expression of the ethylene biosynthetic genes LcACS1/4 and LcACO2/3 in the fruitlet abscission zone(FAZ).Two genes that encode the BR core signaling components brassinazole resistant(BZR)proteins,namely,LcBZR1 and LcBZR2,were characterized.LcBZR1/2 were localized to the nucleus and acted as transcription repressors.Interestingly,the LcBZR1/2 transcript levels were not changed during ET-induced fruitlet abscission,while their expression levels were significantly increased after BR application.Moreover,gel shift and transient expression assays indicated that LcBZR1/2 could suppress the transcription of LcACS1/4 and LcACO2/3 by specifically binding to their promoters.Importantly,ectopic expression of LcBZR1/2 in Arabidopsis significantly delayed floral organ abscission and suppressed ethylene biosynthesis.Collectively,our results suggest that BRs suppress ET-induced fruitlet abscission through LcBZR1/2-controlled expression of genes related to ethylene biosynthesis in litchi.In addition,similar results were observed in the Arabidopsis gain-of-function mutant bzr1-1D,which showed delayed floral organ abscission in parallel with lower expression of ACS/ACO genes and reduced ethylene production,suggesting that the mechanism of BZR-controlled organ abscission via regulation of ethylene biosynthesis might be conserved in Arabidopsis.

Seedless fruits have high consumer appeal and have made seeded varieties obsolete in some crops.In seedless apple varieties,core tissues which normally contain the seed can be unpalatable,reducing the seedless appeal.Apples are accessory fruit with edible flesh derived from hypanthial tissue–a floral tube fused to a compound ovary.Here we show that through suppression of AGAMOUS-like carpel identity genes and hormone induced fruit set,it is possible to generate coreless and therefore seedless apples.Suppression of AGAMOUS-like genes increased petal whorls and fully eliminated carpel development.Treatments with a combination of gibberellin,cytokinin and auxin,rather than single treatments,were required for fruit initiation in these lines.Transcriptomic analysis of agamous RNAi lines suggested conservation of AGAMOUS-dependent gene networks between apple and Arabidopsis.In the absence of all sexual tissues,the developing fruit continues to grow and follow a ripening process similar to that of a regular apple.The coreless phenotype offers a new concept for pipfruit consumers improving convenience and reducing food waste.

An adaptive filter based on Data-Reusing Least-Mean-Square algorithm has been proposed and applied in magnetocardiography (MCG) to suppress ambient noise. Numerical simulation studies indicate that the adaptive filter is a powerful noise suppresser for correlated interferences, especially for those with amplitude changing and time delay.Also the filter has a wide frequency bandwidth. With this filter, the signal-to-noise ratio of an MCG is improved to the intrinsic noise level. The periodic average method for further improvement of the noise level in MCG is also discussed.
