The present experiment was designed to elucidate the effects of the sterilized colloids separated from red soil,latosol and yellow brown soil,which are the typical zonal soils in central-south China, on the adsorption,activity,kinetic parameters and thermal stability of urease enzyme which plays significant role in the transformation of soil nitrogen compounds. Results show that the amount of enzyme adsorbed on the examined soil colloids followed the order: yellow brown soil>red soil=latosol. The residual activities of enzyme in different colloid systems are in the following sequence:red soil>latosol>yellow brown soil. The thermal stability of adsorbed enzyme is higher than that of free enzyme.

The influence of quinclorac (3,7-dichloroquinoline-8-carboxylic acid) on enzyme activities in flooded paddy soils was assessed under laboratory conditions. The enzymes differed markedly in their response to quinclorac. Quinclorac inhibited proteinase, hydrogen peroxidase, phosphorylase, and urease activities.The higher the concentration of quinclorac applied, the more significant the inhibition to these observed activities with a longer time required to recover to the level of the control. However, soils supplemented with quinclorac were nonpersistent for proteinase, phosphorylase and urease as opposed to soils without quinclorac. Dehydrogenase activity was also sensitive to quinclorac. Three soil samples with concentrations of quinclorac higher than 1 μg g-1 soil declined to less than 20% of that in the control. However, the highest dehydrogenase activity (up to 3.28-fold) was detected in soils with 2 μg g-1 soil quinclorac on the 25th day after treatment. Quinclorac had a relatively mild effect on saccharase activity at the concentrations used in this experiment and a stimulatory one on soil respiration when added to soil at normal field concentrations.Nonetheless it was inhibited at higher concentrations in paddy soils. Quinclorac is still relatively safe to the soil ecosystem when applied at a normal concentration (0.67 μg g-1 dried soil) but may have some effects on soil enzymes at higher concentrations.

Biochar has been considered as a potential way to enhance acidophilous plant growth in alkaline soils.However,whether such enhancements are closely linked with biochar pyrolysis temperature and its improvements in rhizospheric soil fertility and microbial activities remains largely unknown.We performed a pot experiment to investigate changes in azalea(Rhodo-dendron)morphology and physiology,as well as its rhizosphere soil chemical and biological properties in a slightly alkaline urban soil under the amendment of biochars that pyrolyzed at three temperatures(i.e.,350,550 and 700°C).Our results showed that the effects of biochars on plant growth and soil properties depended on pyrolysis temperature.In comparison with the non-amended control,350 and 550°C biochars showed significant promotions on the azalea growth in terms of photosynthetic characteristics,biomass,root morphology,and N and P uptake.Whereas,700°C biochar showed an inhibi-tion effect on them.350°C biochar decreased soil pH and increased soil available P and K contents and the activities ofα-glucosidase,N-β-glucosaminidase,phosphatase and sulfatase.In addition,350°C biochar significantly enhanced bacterial 16S rRNA and fungal 18S rRNA gene abundances in the rhizosphere soil of azalea and mycorrhizal infection.Correlation analysis indicated that soil pH,available nutrients and fungal abundance had positive associations with the enhanced plant growth parameters.Therefore,our findings suggest that biochar produced at low temperature could be a feasible strategy for enhancing acidophilous azalea growth and improving urban soil quality.

In semi-arid areas of China,gravel and sand mulch is a farming technique with a long history.In this study,a sample survey was conducted on long term gravel sand mulch observational fields in the Northwest Loess Plateau to determine the effects of long term mulch on soil microbial and soil enzyme activities.We found that after long term gravel-sand mulch,compared with bare ground,soil organic matter,alkali nitrogen,conductivity decreased,while pH and soil moisture increased.Urease,saccharase and catalase decreased with increased mulch thickness,while alkaline phosphatase was reversed.The results of Illumina MiSeq sequencing shows that after gravel-sand mulch,the bacterial and fungal community structure was different from bare land,and the diversity was reduced.Compared with bare land,the bacteria Proteobacteria and Acidobacteria abundance increased with increased thickness,and Actinobacteria was opposite.Also,at the fungal genus level,Fusarium abundance was significantly reduced,and Remersonia was significantly increased,compared with bare land.Redundancy analysis(RDA)revealed that soil environmental factors were important drivers of bacterial community changes.Overall,this study revealed some of the reasons for soil degradation after long term gravel-sand mulch.Therefore,it is recommended that the addition of exogenous soil nutrients after long term gravel-sand can help improve soil quality.

An incubation experiment with soil water content treatments of 0.15 (W1), 0.20 (W2), and 0.40 (W3) g g-1 soil was carried out for two months to investigate the activities of important enzymes involved in C, N, P, and S cycling in a paddy soil from the Taihu Lake region, China, under waterlogged and aerobic conditions. Compared with air-dried soil,waterlogging resulted in a significant decrease (P ≤ 0.05) of fluorescein diacetate (FDA) andβ-D-glucosidase activities,and this effect was enhanced with increasing waterlogging time. Waterlogging also significantly inhibited (P ≤ 0.05)arylsulfatase as well as alkaline and acid phosphatase activities, but did not decrease the activities with the increase in waterlogging time. Short-term waterlogging did not affect urease activity, but prolonged waterlogging decreased it markedly. In contrast, the aerobic incubation (W1 and W2 treatments) significantly increased (P ≤ 0.05) FDA, alkaline phosphatase, and β-D-glucosidase activities. With aerobic treatments the activities of FDA and alkaline phosphatase increased with incubation time, whereas β-D-glucosidase activity decreased. A significant difference (P ≤ 0.05) was usually observed between the W1 and W2 treatments for the activities of FDA as well as alkaline and acid phosphatase;however,β-D-glucosidase and urease were usually not significant (P ≤ 0.05). No activity differences were observed between waterlogging and aerobic incubation for arylsulfatase and urease.

Heavy metal pollution has received increasing attention in recent years mainly because of the public awareness of environmental issues. In this study we have evaluated the effect of cadmium(Cd) on enzymes activity, substrate utilization pattern and diversity of microbial communities in soil spiked with 0, 20, 40, 60, 80, and 100 mg/kg Cd, during 60 d of incubation at 25℃. Enzyme activities determined at 0, 15, 30, 45, and 60 d after heavy metal application(DAA) showed marked declines for various Cd treatments, and up to 60 DAA, 100 mg/kg Cd resulted in 50.1%, 47.4%, and 39.8 % decreases in soil urease, acid phosphatase and dehydrogenase activities,respectively to control. At 60 DAA, substrate utilization pattern of soil microbial communities determined by inoculating Biolog ECO plates indicated that Cd addition had markedly inhibited the functional activity of soil microbial communities and multivariate analysis of sole carbon source utilization showed significantly different utilization patterns for 80 and 100 mg/kg Cd treatments. The structural diversity of soil microbial communities assessed by PCR-DGGE method at 60 DAA, illustrated that DGGE patterns in soil simplified with increasing Cd concentration, and clustering of DGGE profiles for various Cd treatments revealed that they had more than 50% difference with that of control.

To reveal the biological characteristics of urban forest soil and the effects of soil enzyme on soil fertility as well as the correlation between physicochemical properties and enzyme activities,44 urban forest soil profiles in Nanjing were investigated.Basic soil physicochemical properties and enzyme activities were analyzed in the laboratory.Hydrogen peroxidase,dehydrogenase,alkaline phosphatase,and cellulase were determined by potassium permanphosphate dinatrium colorimetry,and anthrone colorimetry,respectively.The result showed that soil pH,organic carbon (C),and total nitrogen (N) had great effects on hydrogen peroxidase,dehydrogenase,and alkaline phosphatase activities in 0-20 cm thick soil.However,pH only had great effect on hydrogen peroxidase,dehydrogenase,and alkaline phosphatase activities in 20-40 cm thick soil.Hydrogen peroxidase,dehydrogenase,and alkaline phosphatase were important biological indicators for the fertility of urban forest soil.Both in 0-20 cm and 20-40 cm soil,soil enzyme system (hydrogen peroxidase,dehydrogenase,alkaline phosphatase,and cellulase) had a close relationship with a combination of physicochemical indicators (pH,organic C,total N,available K,available P,cation exchange capacity (CEC),and microbial biomass carbon(Cmic)).The more soil enzyme activities there were,the higher the fertility of urban forest soil.
