In order to discuss the crystal lattice relaxation in DX center, a new X-ray absorption fine structure (XAFS) analysis, 'capacitance XAFS', is applied to the Al_(0.33)Ga_(0.67)As : Se system. Incapacitance XAFS, the X-ray absorption of a defect atom with a localized electron is selectively observed by measuring the photon-energy dependence of the capacitance. Capacitance XAFS at the Ga K-edge is different from conventional XAFS, although a similar absorption spectra are obtained at the As K-edge, suggesting that the DX center in this system originates from the trigonal broken bond (BB-DX) configuration rather than the cation-cation bonded (CCB-DX) state.

The Magneto-Inertial Fusion (MIF) approach has the benefits of both approaches of Inertial Confinement Fusion (ICF) and Magnetic Confinement Fusion (MCF). MIF is a complete approach, as compared to others. It has the advancement of the common ICF and some advantages over MCF. It also includes high-density plasma in the presence of strong magnetic fields. In this study, we investigate the inverse bremsstrahlung absorption (IBA) in magnetized plasma and the kinetic theory. Many researchers have calculated the inverse bremsstrahlung absorption coefficient through the ICF approach. In this investigation, however, the average of IBA is obtained that can be used in the MIF approach, the kinetic theory and the Krook collision frequency. In addition, the influences of some parameters such as laser wavelength, electron temperature, and static magnetic field are shown on the mean collisional absorption. A numerical solution is also used to calculate the dispersion function and the laser electric field. IBA is one of the basic heating processes in laser fusion plasma. The results of the study show that, in critical layers, the increase of absorption is proportional to short laser wavelength, low electron temperature, but IBA varies slowly with the static magnetic field.

Purpose The angiotensin converting enzyme-2 (ACE2)—entry receptor of SARS-CoV-2—and its homologue, the angiotensin-converting enzyme (ACE), play a pivotal role in maintaining cardiovascular homeostasis. Potential changes in ACE2 expression levels and dynamics after SARS-CoV-2 infection have been barely investigated. The aim of this study was to develop an ACE2-targeting imaging agent as a noninvasive imaging tool to determine ACE2 regulation. Methods DOTA-DX600, NODAGA-DX600 and HBED-CC-DX600 were obtained through custom synthesis and labeled with gallium-67 (T1/2?=?3.26 d) as a surrogate radioisotope for gallium-68 (T1/2?=?68 min). ACE2- and ACE-transfected HEK cells were used for the in vitro evaluation of these radiopeptides. The in vivo tissue distribution profiles of the radiopeptides were assessed in HEK-ACE2 and HEK-ACE xenografted mice and imaging studies were performed using SPECT/CT. Results The highest molar activity was obtained for [67Ga]Ga-HBED-CC-DX600 (60 MBq/nmol), whereas the labeling efficiency of the other peptides was considerably lower (20 MBq/nmol). The radiopeptides were stable over 24 h in saline (>?99% intact peptide). All radiopeptides showed uptake in HEK-ACE2 cells (36–43%) with moderate ACE2-binding affinity (KD value: 83–113 nM), but no uptake in HEK-ACE cells (<?0.1%) was observed. Accumulation of the radiopeptides was observed in HEK-ACE2 xenografts (11–16% IA/g) at 3 h after injection, but only background signals were seen in HEK-ACE xenografts (<?0.5% IA/g). Renal retention was still high 3 h after injection of [67Ga]Ga-DOTA-DX600 and [67Ga]Ga-NODAGA-DX600 (~?24% IA/g), but much lower for [67Ga]Ga-HBED-CC-DX600 (7.2?±?2.2% IA/g). SPECT/CT imaging studies confirmed the most favorable target-to-nontarget ratio for [67Ga]Ga-HBED-CC-DX600. Conclusions This study demonstrated ACE2 selectivity for all radiopeptides. [67Ga]Ga-HBED-CC-DX600 was revealed as the most promising candidate due to its favorable tissue distribution profile. Importantly, the HBED-CC chelator enabled 67Ga-labeling at high molar activity, which would be essential to obtain images with high signal-to-background contrast to detect (patho)physiological ACE2 expression levels in patients.

While aminoacyl-tRNA synthetase-interacting multifunctional protein 2 (AIMP2) is a tumor suppressor, its exon 2-depleted splice variant (AIMP2-DX2 or shortly DX2) is highly expressed in human lung cancer, and the ratio of DX2 to AIMP2 increases according to the progression of lung cancer. In this study, pyrimethamine inhibited the level of DX2 (IC50 = 0.73 μM) in A549 cells expressing nanoluciferase-tagged DX2. In a panel of 5 lung cancer cell lines with various DX2 levels, pyrimethamine most potently suppressed the growth of H460 cells, which express high levels of DX2 (GI50 = 0.01 μM). An immunoblot assay in H460 cells showed that pyrimethamine decreased the DX2 level dose-dependently but did not affect the AIMP2 level. Further experiments confirmed that pyrimethamine resulted in ubiquitination-mediated DX2 degradation. In an in vivo mouse xenograft assay using H460 cells, intraperitoneal administration of pyrimethamine significantly reduced the tumor size and weight, comparable with the effects of taxol, without affecting body weight. Analysis of tumor tissue showed a considerably high concentration of pyrimethamine with a decreased levels of DX2. These results suggest that pyrimethamine, currently used as anti-parasite drug, could be repurposed to treat lung cancer patients expressing high level of DX2.

The developed dynamic model of the main geomagnetic field (MGF) includes the sources of three levels, which have existed and continuously changed during the 100-year period. The spatial-temporal dynamics of the secular variations in MGF has been considered based on this model. It has been indicated that, at different times, the formation of anomalies (located in similar regions) depended on the total dynamics of different combinations of sources, including a change in the parameters of the dipoles of the secondorder smallness. Anomalies generated by the dynamics of the dipoles of the third-order smallness do not evidently manifest themselves in the total secular variations but are responsible for the specific shapes of isodynamic lines. Based on the stability and continuity of the constructed model, the conclusion is made that long-living turbulent flows of different scales can exist in the liquid core.
