Staphylococcus aureus is a leading cause of both acute and chronic infections in humans. The importance of the pentose phosphate pathway (PPP) during S. aureus infection is currently largely unexplored. In the current study, we focused on one key PPP enzyme, transketolase (TKT). We showed that inactivation of the unique gene encoding TKT activity in S. aureus USA300 (tkt) led to drastic metabolomic changes. Using time-lapse video imaging and mice infection, we observed a major defect of the tkt strain compared with wild-type strain in early intracellular proliferation and in the ability to colonize kidneys. Transcriptional activity of the 2 master regulators sigma B and RpiRc was drastically reduced in the tkt mutant during host cells invasion. The concomitant increased RNAIII transcription suggests that TKT-or a functional PPP-strongly influences the ability of S. aureus to proliferate within host cells by modulating key transcriptional regulators.

Objectives Pericardial involvement is a frequent manifestation of systemic lupus erythematosus (SLE). Growing evidence suggests that colchicine may be useful for acute or recurrent pericarditis. We report for the first time a series of 10 consecutive cases of SLE with pericarditis treated with colchicine.Methods Inclusion criteria in this retrospective study were diagnosis of SLE, pericarditis and receiving colchicine.Results We included 10 consecutive cases of SLE with pericarditis treated with colchicine (nine women, mean age at the index pericarditis 3512 years). Pericarditis was the initial manifestation of SLE for two patients, whereas eight patients had SLE lasting for a median of 2.5 years (15 days to 13 years) and had received prednisone (n=7, 2-30mg/d), hydroxychloroquine (n=7), azathioprine (n=3), methotrexate (n=2), and mycophenolate mofetil (n=1). For six patients, pericarditis was associated with other SLE manifestations. Altogether, colchicine avoided the use (n=2) or increase in dosage (n=5) of steroids in seven cases; the increase in steroids dosage was minimal for two patients. Colchicine 1mg was given for a median of 39 days (10 days to 54 months). Symptoms completely resolved after a median of 2.5 days (1-30 days) after initiation of colchicine. Colchicine was maintained or resumed in six patients to prevent recurrence, with no further relapse. Conclusions Colchicine may be safe and effective in treating SLE pericarditis and used as a steroids-sparing agent. These preliminary results need to be confirmed in a larger study with longer follow-up.

Healthcare workers have been facing the COVID-19 pandemic, with numerous critical patients and deaths, and high workloads. Quality of care is related to the mental status of healthcare workers. This PRISMA systematic review and meta-analysis, on Pubmed/Psycinfo up to October 8, 2020, estimates the prevalence of mental health problems among healthcare workers during this pandemic. The systematic review included 70 studies (101 017 participants) and only high-quality studies were included in the meta-analysis. The following pooled prevalences were estimated: 300 % of anxiety (95 %CI, 24.2-37.05); 311 % of depression (95 %CI, 25.7-36.8); 565 % of acute stress (95 %CI - 30.6-80.5); 20,2% of post-traumatic stress (95 %CI, 9.9-33.0); 44.0 % of sleep disorders (95 %CI, 24.6-64.5). The following factors were found to be sources of heterogeneity in subgroups and metaregressions analysis: proportion of female, nurses, and location. Targeted prevention and support strategies are needed now, and early in case of future health crises.

The analysis of urinary protein composition is an important step in the evaluation and monitoring of kidney diseases. Among the various approaches, the determination of urinary-specific proteins makes it possible to non-invasively detect a preferentially tubular or glomerular injury, to orientate towards a pathological process, to guide the indication of a kidney biopsy, and to follow the evolution of the disease and the effectiveness of a therapy. No study systematically evaluated the performance of urinary-specific proteins for the diagnosis of a renal disease. We conducted this retrospective study to perform an exhaustive analysis of the correlations that may exist between histologically proven kidney disease and the corresponding specific urinary protein composition it in order to evaluate the diagnostic value of each of its components. Urinary concentrations of total protein, albumin, transferrin, alpha1microglobulin, beta2microglobulin, retinol binding protein, and immunoglobulin G were analyzed in more than 500 patients who underwent renal biopsy and concomitant urine specific protein analysis. Our analysis shows that these markers have a limited positive predictive value in this cohort of complex and unselected kidney diseases. In particular, low molecular weight proteins, and especially alpha1microglobulin, are frequently associated with glomerular diseases. We identified transferrin as an independent predictor of minimal changes disese and renal amyloidosis, and beta2microglobulin as an independent predictor of acute tubulointerstitial nephropathy and myelomatous tubulopathy. Finally, we defined the thresholds at which these parameters had excellent negative predictive values.

Following a maternal primary infection in the first trimester, up to 30% of infected neonates suffer long-term sequelae. Maternal parity is an established risk factor of cCMV in previously seronegative women. Our objective was to quantify, in a population of women seronegative at their first pregnancy, the risk of cCMV and related sequelae following primary infections in the first trimester in subsequent pregnancies.
