An amperometric glucose sensor was constructed using glucose oxid... 展开 A new method of physically immobilizing a biomolecule of analytical interest in poly(vinyl alcohol) cryogels was developed for use in the manufacture of biosensors. An amperometric glucose sensor was constructed using glucose oxidase immobilized onmembranes obtained by a cyclic freezing/thawing process. No chemical crosslinking agent was used. Sensor behaviour was evaluated electrochemically with a hydrogen peroxide electrode. When used to determine glucose in standard solutions, the sensor gave linear calibration curves in the range 0.05-3 mM. Temperature and pH effects on the electrochemical response are reported. Kinetic parameters indicated that the immobilized glucose oxidase behaved almost like the free enzyme, but that enzyme leakage was considerable. 收起
